Ku protein

Ku is a dimeric protein complex that binds to DNA double-strand break ends and is required for the non-homologous end joining (NHEJ) pathway of DNA repair. Ku is evolutionarily conserved from bacteria to humans. The ancestral bacterial Ku is a homodimer (two copies of the same protein bound to each other). [2] .

It comprises the DNA-PK catalytic subunit (DNA-PKcs) and the heterodimer of DNA-binding proteins Ku70 and Ku80. Here, we report the cryo-electron microscopy (cryo-EM) …Here, small angle X-ray scattering (SAXS) and mutational analyses show APLF is largely an intrinsically disordered protein that binds Ku, Ku/DNA-PKcsmore » X4L4 assembles with Ku heterodimers linked to DNA-PKcs via flexible Ku80 C-terminal regions (Ku80CTR) in a complex stabilized through APLF interactions with Ku, DNA-PK, and X4L4.

Human Ku70 is a well-known endogenous nuclear protein involved in the non-homologous end joining pathway to repair double-stranded breaks in DNA. However, Ku70 has been studied in multiple contexts and grown into a multifunctional protein. In addition to the extensive functional study of Ku70 in DNA repair process, many studies have emphasized the role of Ku70 in various other cellular ...Proteins are made up of amino acids. All proteins made in living organisms consist of combinations of 20 amino acids. These contain carbon, hydrogen, oxygen, nitrogen and sometimes sulfur and phosphorus.Non-homologous end joining proteins and their known interactions. a | The non-homologous end joining (NHEJ) DNA protein kinase (DNA-PK) complex consists of a heterodimer of Ku70 and Ku80 plus DNA-PK catalytic subunit (DNA-PKcs). Ku70 and Ku80 consist of von Willebrand (vWA) domains, the Ku core and the nuclear localization sequence (NLS).Ku, another protein required for NHEJ, also stimulates LX ligation non-specifically and enhances DNA bridging [17]. Indeed, there is a striking similarity between Ku and AHNAK in this context since Ku also interacts with LX on the DNA molecule and, based on a specific interaction with LX, has been suggested to recruit LX to DNA [53] .

Ku (Ku70/Ku80) is a DNA-binding protein involved in DNA repair, playing a fundamental role for the repair of double-stranded DNA breaks through the non-homologous end-joining (NHEJ) pathway [4], [5]. The Ku complex can be responsible for a very strong autoimmune answer in autoimmune disease and anti-Ku antibodies may have significant impact on ...Ku protein binds to DNA ends and is a cofactor for the DNA-dependent protein kinase. Both of these components are involved in DNA double-strand break repair, but it has not been clear if they function indirectly, by sensing DNA damage and activating other factors, or if they are more directly involved in the processing and rejoining of DNA breaks.NHEJ requires a protein complex consisting of the heterodimer of Ku70 and Ku80 (Ku complex) that recognizes DSB free ends and the DNA-dependent protein kinase catalytic subunit (DNA-PKcs) (1, 5, 6) that recruits core NHEJ factors including Artemis, XRCC4, Ligase IV, and XLF on DSB break sites ().DNA-PKcs is a ∼470-kDa serine-threonine kinase belonging to the phosphoinositide 3-kinase-related ...Aug 15, 1999 · Ku protein binds free DNA ends, recruiting the DNA-PK. On interaction with an opposing DSB-DNA-PK complex, transphosphorylation occurs between DNA-PKs, leading to the activation of Ku-helicase, allowing microhomology base-pairing. Finally, unpaired DNA is resolved, and DNA ends are ligated. This results in characteristic …Ku bound at a DNA end then recruits the protein PAXX (refs 5, 6) plus the catalytic subunit of the DNA-dependent protein kinase (DNA-PKcs) 7. The complex of Ku and DNA-PKcs at the DNA end forms ...

Two distinct and complementary pathways, non-homologous end joining (NHEJ) and homologous recombination (HR), are employed by mammalian cells to repair DNA DSBs. Each pathway is tightly controlled in response to increased DSBs. The Ku heterodimer has been shown to play a regulatory role in NHEJ repair. Ku80 ubiquitination contributes to the ...KU80; Ku-core domain, Ku80 subfamily; Ku80 is a subunit of the Ku protein, which plays a key role in multiple nuclear processes such as DNA repair, chromosome maintenance, transcription regulation, and V (D)J recombination. This means that a 1 mg/ml solution of the protein will have an absorbance at 280 nm of 0.54. E (0.1%, 280) can be used as a very convenient measure of the protein content provided that the enzyme preparation supplied by Sigma is pure. A 'rule of thumb', useful when the E (0.1%, 280) is unknown, is that a 1mg/ml protein solution has an A280 of 1.In contrast, PHR 2 and 3 ( Gell and Jackson 1999) map to the amino-terminal region exclusively shared by the eukaryotic Ku proteins. This core domain shared by the prokaryotic YkoV-like proteins and the eukaryotic Ku70 and Ku80 (hereinafter Ku core; Fig. 2) is ~234-280 amino acids long and is larger than most common globular domains.Nov 13, 2020 · with identification of the Ku protein as an autoantigen in sclero-derma-polymyositis overlap syndrome (9, 10). In the hope of gain-ing insight into the pathogenesis of autoimmune disease, Ku was extensively characterized. Ku is a heterodimer of 70 and 86 kDa (Ku70 and Ku86) that binds with strong affinity to DNA ends, stem-loop or bubble struc-

Ku protein, together with Rap1p, a telomeric DNA (TG 1-3 repeat)-binding protein, is also required to initiate transcriptional silencing, or telomere-position effect (TPE).
