The way I read enlarge/reduce is it's effectively two spells in one prepared place. So you cast enlarge, and can apply twin metamagic to enlarge two targets. Or you cast reduce, and can apply twin metamagic to shrink two targets. It's still just the one casting of the spell, you've just incremented the number of targets from 1 to 2..
